ABSTRACT

The leucine repeat-rich G-protein-coupled receptor 5 (Lgr5), which expressed at the bottom of the crypt, not only acts as a marker of intestinal stem cells, but also induces cytonemes for transfering signaling effectors by internalization, influences proliferation by Wnt /β-catenin pathway, and promotes cytoskeletal structure, cell adhesion of intestinal stem cells(ISCs) via the IQGAP1. In addition, Lgr5 can be affected by the regulation of IL-1β、IL-22、miR-100-5p and others. Besides, intestinal microbiota change the localization of Lgr5+ cells in cript to induce apoptosis increased. Therefore,Lgr5 is influenced by many factors, which may play an important role in the proliferation, differentiation and stability of ISCs.

ABSTRACT

<p><b>OBJECTIVE</b>To Study the effect of C677T and MTHFR gene polymorphism on side effects of HD-MTX in ALL children.</p><p><b>METHODS</b>The gene polymorphism of C677T A303G and MTHFR C677T were detected by PCR in 98 ALL children from January 2014 to January 2016. The side effects during HD-MTX therapy were observed, and the relationship among GSTP1, MTHFR gene polymorphism and incidence of side effect of HD-MTX were analyzed.</p><p><b>RESULTS</b>Among 98 ALL children, the gene variation was observed in 61 ALL children (62.24%). Polymorphism study on C677T A303G showed that the gene frequency of A was 84.69%, while that of G was 15.31%; for polymorphism of MTHFR C677T, gene frequency of C was 66.33%, and that of T was 33.67%. Seven patients(7.14%) experienced with bone marrow supression, 23 patients(23.47%) with liver function damage, 15 patients(15.31%) with renal function damage, 48 patients(48.98%) with gastrointestinal reactions and 46 patients(46.94%) with mucosal lesions. After adjustment of sex, age, risk stratification and dosage of MTX, the gene polymorphism had no significant relationship with bone marrow suppression, gastrointestinal reactions and mucosal lesions(P>0.05). However, the number of the mutant genes had statistically significant relationship with liver and renal function damage(P<0.05).</p><p><b>CONCLUSION</b>The risk of side effects during HD-MTX therapy increases in ALL children with combined mutation of MTHFR and C677T.</p>

ABSTRACT

Objective To determine the hyperelastic parameters shear modulus (μ) and curvature parameter (α) of extraocular muscles (EOMs) in Ogden hyperelastic model, so as to provide theoretical basis for clinical EOM surgery by numerical modeling. Methods The passive behavior of fox EOMs in vitro was determined by the uniaxial tensile test, and the hyperelastic analysis was conducted by the first-order Ogden model and ABAQUS software. Results The experimental result showed that the passive behavior of fox EOMs was nonlinear. The corresponding hyperelastic parameters μ =(6.57±3.76) kPa and α=8.16±1.63 were obtained. When the strain of EOMs was larger than 6%, there were no statistical differences between the experimental result and the calculation result of the first-order Ogden hyperelastic model (P>0.05). Both the calculation result and the simulation result well fitted to the experimental result. Conclusions The hyperelastic parameters identified in this study can be used as the input for the corresponding numerical modeling of fox EOMs.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effects of 17beta-estradiol on the intracellular calcium of masticatory muscles myoblast.</p><p><b>METHODS</b>Myoblasts from maxillofacial skeletal muscle of one week old female Sprague-Dawley rats were cultured. Fluo-4-AM as the Ca2+ indicator and the laser confocal microscope system were used to observe the effects of estrogen on the cytoplasmic Ca2+ concentration in the normal pH condition and the acid condition (pH = 6.7).</p><p><b>RESULTS</b>In the normal pH condition, when 17beta-estradiol (10(-9), 10(-8), 10(-7) mol/L) were added to cells cytoplasmic Ca2+ immediately increased then decreased right away, and in the end came into a new Ca2+ homeostasis in the base line. In the acid condition, 17beta-estradiol (10(-9), 10(-8), 10(-7) mol/L) made the cytoplasmic Ca2+ decreased immediately then came into a new Ca2+ homeostasis under the base line.</p><p><b>CONCLUSION</b>The results suggest that estrogen may maintain the skeletal cytoplasmic Ca2+ concentration in a lower level and reduce the cytoplasmic Ca2+ accumulation to keep the normal functions of masticatory muscles myoblast.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To localize the gene (s) responsible for autosomal dominant hypocalcified amelogenesis imperfecta in a Chinese family.</p><p><b>METHODS</b>A Chinese family which was diagnosed as autosomal dominant hypocalcified amelogenesis imperfecta (AD) was studied. Venous blood from nineteen family members was collected and genomic DNA was extracted from the blood. Eight short tandem repeats (STRs) spanning five hereditary AI candidate genes were selected and linkage analysis between the genetic markers and the disease loci was performed.</p><p><b>RESULTS</b>Genotype of the eight STRs were acquired, the linkage analysis result can not support that the gene for AI pedigrees was linked to ENAM, AMBN, TUF1, KLK4 or MMP-20.</p><p><b>CONCLUSION</b>The results can not support all proposed candidate gene regions as causal for autosomal dominant hypocalcified AI in this family. These linkage findings provide further evidence for genetic heterogeneity among families with autosomal dominant AI and indicate that, at least, some forms of autosomal dominant AI are not caused by a gene in the five most commonly reported AI candidate genes.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the antibody responses induced by recombinant plasmid plRES-fimA:IL15 via nasal immunization to BABL/c mice and the regulation of IL-15 to sIgA.</p><p><b>METHODS</b>BABL/c mice were immunized with recombinant plasmids pIRES-fimA:IL15 and pIRES-fimA via nasal or intramuscular route. Serum IgG and salivary sIgA levels after immunization were analyzed by ELISA.</p><p><b>RESULTS</b>Nasal immunization with plasmids pIRESfimA:IL15 or pIRES-fimA elicited significant higher level of salivary FimA-specific sIgA responses compared with intramuscular immunization. There was no significant difference of the serum IgG responses between nasal immunization mice and intramuscular immunization mice. Nasal immunization with plasmid pIRES -fimA:IL15 elicited significant higher level of salivary sIgA response than with pIRES-fimA (P<0.05).</p><p><b>CONCLUSION</b>Nasal dropping may be an effective mucosal immunization route of anti-Porphyromonas gingivalis DNA vaccine to elicit specific antibody responses in serum and oral region. IL-15 has a positive regulation effect to sIgA response.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore an effective therapy for treatment of infantile febrile convulsion.</p><p><b>METHODS</b>Seventy infants of febrile convulsion were randomly divided into two groups, a combined acupuncture and drug group (n = 36) were treated with Xingnao Kaiqiao needling method with Shuigou (GV 26) selected as main combined with intramuscular injection of luminal and intravenous perfusion of diazepam; and a drug group (n = 34) treated with simple intramuscular injection of luminal and intravenous perfusion of diazepam. Their therapeutic effects, the time of inducing the stopping convulsion, and the recurrence rate were investigated.</p><p><b>RESULTS</b>The total effective rate was 86.1% in the combined acupuncture and drug group, better than 79.4% in the drug group (P<0.05), and the time of inducing the stopping convulsion in the combined acupuncture and drug group was shorter than that in the drug group (P<0.01), and the recurrence rate in the combined group was lower than that in the drug group (P<0.05).</p><p><b>CONCLUSION</b>Acupuncture combined with intramuscular injection of luminal and intravenous perfusion of diazepam is an effective and safe method for febrile convulsion.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To construct a eukaryotic co-expression plasmid pIRES-fimA:IL15, which can be used as an immunoreaction-enhancing DNA vaccine against Porphyromonas gingivalis FimA, and investigate its expression in mammalian cells.</p><p><b>METHODS</b>The eukaryotic co-expression plasmid pIRES-fimA:IL15 was constructed by molecular cloning methods and characterized by restricted endonuclease mapping, PCR and DNA sequencing. The plasmid was transfected into mammalian cell CHO using Lipofectamine 2000. Expression of fimA gene was detected by Western blot and the protein secretion in cultural medium was analyzed by ELISA.</p><p><b>RESULTS</b>Endonuclease mapping showed that the target genes fimA and IL-15 obtained by PCR had the same molecular size as predicted. The DNA sequencing data also indicated that inserted fimA gene and IL-15 gene had correct DNA sequence and orientation. The recombined plasmid could express FimA in mammalian cell CHO transfected. FimA and IL-15 could be secreted into cultural supernatant detected by ELISA.</p><p><b>CONCLUSION</b>A new eukaryotic co-expression plasmid pIRES-fimA: IL15 was constructed and it could be applied for further immunization in animal as an effective anti-Porphyromonas gingivalis vaccine.</p>

ABSTRACT

Objective To explore the relationship between tumor necrosis factor(TNF)-? promoter G238A,G308A and interferon(IFN)-?+874A/T polymorphism and susceptibility to cytomegalovirus(CMV) hepatitis.Methods A TaqMan fluorescence polymerase chain reaction(PCR) in the IFN-?+874A/T and TNF-? gene promoter region single nucleotide polymorphism was tested in the subjects,including 87 infants with CMV hepatitis and 89 children without CMV hepatitis.Results No evident difference of TNF-? +238G/A and+308G/A allele frequency was found between infants with CMV hepatitis and controls.The cases of IFN-?+874 AA,AT and TT genotype were 64,20,3 cases in the infant CMV hepatitis group,and 45,26 and 18 cases in the control group,respectively.There was significant difference in IFN-?+874 allele genotype and frequency between infants with CMV hepatitis and the control group(P=0.001,P

ABSTRACT

Objective To investigate the relationship between cytomegalovirus infection and biliary atresia.Methods Sixteen infants with CMV infection and biliary atresia were retrospectively reviewed and compared with clinical manifestations with 29 infants with CMV hepatitis.Results Both serum CMV-IgM antibody and CMV-pp65 in polymophnuclear leukocytes were found positive in 9 cases,only CMV IgM antibody was found positive in 1 case and only CMV-pp65 were found positive in 3 cases,both CMV-IgM and CMV-pp65 were found negative in 3 cases.14 of 15 cases of CMV-pp65 were detected in liver tissues including 3 cases with negative CMV-IgM antibody and CMV-pp65 in blood.The clinical indexes in these 16 cases were much more severe than which in 29 infants with CMV hepatitis (P
